According to GMP rules, in aseptic operation zones microbiological monitoring must be performed frequently using a combination of methods, including settle plates, volumetric air sampling, glove and surface sampling.
Passive sampling in combination with active sampling is recognized by regulatory organizations as useful for assessing microbial quality of air environment. During passive sampling or surface sampling, air samplers are not required. However, there are special tools for standardizing operations.
The sedimentation method is applied with a culture plate in horizontal or inclined position depending on the direction of unidirectional airflow. In horizontal position, airflow can be obstructed, creating slight turbulence over agar surface, which can deflect viable particles from agar surface. Inclined position of Petri dish allows air to impact agar surface and pass without disturbances.
For passive sampling, operator can use SETTLE TABLE, fabricated from stainless steel and allowing precise determination of open plate placement location for specified time. After exposure, plate is incubated by standard method. Recommended medium — Tryptic Soy Agar (TSA). Maximum exposure time — 4 hours (90 mm Petri dishes) according to WHO guidelines.
Settle plate exposure in unidirectional airflow conditions
TRIO.SETTLE FOR TABLE is an adjustable device for standardization, testing and counting of microorganisms during passive air monitoring by settle plate method. Passive sampling provides quantitative analysis of microorganisms deposited on agar plate surface during exposure period. Can be mounted on tripod or other support devices. Fabricated from AISI 316 stainless steel, fully autoclavable. Validated according to EN 17141.
